My mom - a house projects maven - pointed me to Soft Coverings when I was looking for cost-effective sources for upholstery for two chairs I was working on.
Located on the top of Queen Anne, this store offers the perfect combination of great selection, good prices, and most importantly, good quality. If you've ever been into a JoAnn Fabrics or Calico Corners, you'll know what I mean - rather than wading through rooms full of polyester florals, Soft Coverings has edited its inventory to include only the kinds of materials and designs I'd want in my own house - making the selection process much easier.
The staff there are also great - great at answering questions, helpful to the relative novice (like myself), but also knowledgeable enough to help more experienced buyers (like my mom) find the perfect fabric.

This store has a huge variety of outdoor and sporting fabrics like fleece, gore-tex, nylon, canvas, mesh and cordura plus all the accouterments you might need to make your own recreational equipment. I have gone here to find replacement buckles, nylon fabric to make a new ground cloth for my tent, canvas for some beach bags and fleece for kiddie hats. The staff is knowledgable and generally friendly and they have all the materials you might need for your project including the patterns. Who knows, you just might walk out with everything you need to make your own three season tent!

More
I end up at the Acorn Street Yarn Shop a few times a year for various craft projects I suddenly and haphazardly undertake. I go in for embroidery floss, fasteners, tools & the like - I'm no knitter. Therefore my impression that they have a boat load of yarn options available is probably obvious to some. Service has been nothing but cheery and sincere.
In general I drive to this store over others because the minute I walk in the door I feel like I'm being hugged by an intimate group of older (than myself) women united in their enthusiasm for domestic crafts. They're knowledgable, friendly, and most of all passionate about their hobby. It's endearing and contagious.

More
Gargoyles Statuary is a unique store that makes shopping almost seem fun. The store is a repository for gothic statues and mystical figurines. They also have an assortment of candles and other home accessories available for sale. While many items in this store are not cheap, the prices overall are not bad. There are some nice pieces for under twenty dollars and of course the accessories are not expensive. The store has a website and catalog for those from out of town.

This place has some of the strongest herbal and tribal candles i have ever smelled. I can even smell the powerful scents from outside of the door. Adding to the ambience, they have plenty of tribal music playing loudly on their speakers, along with a wishing wheel outside. Though it is dented in on one side im sure the wishes you make on it will still come true. Fun place if you like weird stuff.

Hancock fabric has a great selection of quilting fabric and tons of. They also have a good range of decorator fabrics. Sewing and needlework accessories (including yarn, embroidery thread, and knitting needles) are also available. I was impressed by how many special occassion fabrics they had, though they seemed a bit out of date. Lots of flannel and a few seasonal apparel fabrics. Some patterns, but not a huge selection.

It might not be appropriate for a grandmother-type, but Stiches is a new independly-owned fabric store on Capitol Hill. The website trumpets the selection of stretch vinyls, but they also sell lovely cotton prints and nice silks and brocades. They also have a great selection of vintage-style prints.

More
Display and Costume Supply is a costume and party supply store near the Northgate Mall. They carry a wide variety of costumes, from simple rubber masks and makeup to elaborate full body clothing costumes. If you can think of a party theme, they probably have plates, favors, and decorations in stock to match your desires. Employees are generally helpful throughout the store and usually courteous. Prices run a wide range from quite reasonable to unbelievably outrageous. The store has plenty of parking and excellent hours. There aren't too many other stores like it around town, so even if it is pricey, it may be your only bet unless you want to chance things on the internet.

More
They have everything you could imagine for beginners and experts. Their book selection is extensive and their class schedule is truly impressive. What really sets the store apart is their monsterous fabric selection, which is a stash-builder's dream.
They have come out with their own line of fabric which tends toward the flowery side of things, but you can find alternatives on the shelves as well.
Unfortunately, their location is a little challenging. On Lake City Way and a tiny garage with skinny spaces trying to squeeze an SUV in is almost laughable. None the less, if you're a quilter and anywhere near it you need to stop.
